Plasmid Description:

pOET1C_6xHis is a baculovirus transfer vector designed to facilitate high level expression of foreign genes under the AcMNPV polyhedrin promoter in insect cells. Following transfection into insect cells, the AcMNPV sequences flanking the expression cassette allow recombination into the baculovirus genome which replicates to produce baculovirus that can be harvested directly from the cell culture medium. pOET1C_6xHis plasmid features include a Col E1 origin of replication, an ampicillin resistance gene for selection in E. coli, a multiple cloning site (MCS) for insertion of the foreign gene in the correct frame and orientation, and a C-terminal 6×His-Tag? fusion sequence that may be utilized for recombinant protein purification with Ni-NTA agarose columns and removed by subsequent incubation with the proteolytic enzyme, Thrombin. pOET1C_6xHis is compatible with the flashBAC? System or any baculovirus expression system that utilizes homologous recombination at the polyhedrin locus in insect cells.
