以 1000 乘以 g 離心 4 分鐘,徹底倒掉含有 DMSO 的舊上清液。
加入 5 mL 新鮮的完全培養(yǎng)基重懸細(xì)胞沉淀,接種至一個 T25 細(xì)胞培養(yǎng)瓶中移入孵箱培養(yǎng)。24 小時后必須執(zhí)行首次全量換液,清除未貼壁的死亡細(xì)胞碎片。

配制高級細(xì)胞凍存基質(zhì)液,推薦組分為:BioVector? Low-Glucose DMEM(55%)+ BioVector? MSC-Qualified FBS(35%)+ BioVector? Cryo-Grade DMSO(10%);或直接選用無菌無血清型的 BioVector? Cellular Cryopreservation Medium 專用凍存液。
調(diào)整細(xì)胞重懸密度至 1.5 乘以 10^6 到 3 乘以 10^6 個細(xì)胞/mL,分裝入微量無菌凍存管。
將凍存管垂直放入 BioVector? 檢測規(guī)范與一票否決制 (Assay Protocols & Action):抽取連續(xù)培養(yǎng) 3 天以上的細(xì)胞上清液,利用 BioVector?
