BioVector? Primary Normal Human Epidermal Keratinocytes (NHEK) are cells isolated directly from human skin tissue (typically neonatal foreskin or adult surgical specimens). Keratinocytes are the predominant cell type in the epidermis, accounting for over 90% of the total epidermal cell population. In vitro, these cells retain their native biological characteristics, including proliferative capacity and the potential to differentiate into the cornified layer. They serve as a fundamental cellular model for studying skin barrier function, wound healing, inflammatory responses, skin toxicology, and cosmetic efficacy testing.

生物安全性: 所有供體組織均經(jīng)過 HIV, HBV, HCV 等病毒檢測,結(jié)果均為陰性。
2. 培養(yǎng)條件
基礎(chǔ)培養(yǎng)基: BioVector? 角質(zhì)形成細(xì)胞專用無血清培養(yǎng)基 (Keratinocyte Growth Medium, KGM)
關(guān)鍵添加劑: BPE (牛垂體提取物)、EGF (表皮生長因子)、胰島素、氫化可的松。
培養(yǎng)環(huán)境: 37 攝氏度,5% CO2
傳代比例: 1:2 至 1:4;建議在細(xì)胞匯合度達(dá)到 70% 至 80% 時傳代。注意: 原代細(xì)胞不建議培養(yǎng)至 100% 匯合,否則會導(dǎo)致不可逆的分化。
3. 細(xì)胞應(yīng)用
皮膚替代模型: 構(gòu)建 3D 有組織表皮模型 (RHE)。

4. 注意事項

有限壽命: 原代細(xì)胞具有有限的倍增次數(shù)(通??蓚?3 至 5 代),隨著代數(shù)增加,細(xì)胞會逐漸衰老。

2. Culture Conditions
Basal Medium: BioVector? Keratinocyte Growth Medium (KGM, Serum-Free)

Incubation: 37 degrees Celsius, 5% CO2
Subculturing: 1:2 to 1:4 ratio; subculture at 70% to 80% confluence. Crucial: Never allow primary keratinocytes to reach 100% confluence as this triggers irreversible terminal differentiation.

4. Key Usage Notes
Calcium Sensitivity: Keratinocyte behavior is strictly regulated by calcium levels. Low calcium concentrations promote proliferation, while higher concentrations (e.g., above 0.1 mM) induce differentiation.
Finite Lifespan: As primary cells, NHEKs have a limited doubling capacity (typically 3 to 5 passages). Cellular senescence increases with each passage.
Note: To maintain the undifferentiated, proliferative state of BioVector? NHEK cells, it is essential to use BioVector? specialized Serum-Free Medium and minimize exposure to proteolytic enzymes during subculturing. Handle cells with extreme care to ensure high attachment efficiency post-thaw.
