BioVector? MMNK-1是一種高度分化的永生化人膽管上皮細(xì)胞株。

• 來源： 人胎兒肝臟（OUMS-21）。

• 細(xì)胞類型： 永生化正常膽管細(xì)胞。

• 永生化方法： 通過轉(zhuǎn)導(dǎo) SV40 大T抗原 (SV40T) 和人端粒酶逆轉(zhuǎn)錄酶 (hTERT) 實現(xiàn)。

• 形態(tài)： 上皮樣（epithelial-like）。

• 細(xì)胞標(biāo)志物： 表達(dá)膽管細(xì)胞特異性標(biāo)志物，包括 細(xì)胞角蛋白-7 (CK-7) 和 細(xì)胞角蛋白-19 (CK-19)。

• 功能特性：

  • 在 Matrigel 實驗中可形成膽管樣管狀結(jié)構(gòu)。

  • 經(jīng)脂多糖 (LPS) 處理后產(chǎn)生 白細(xì)胞介素-6 (IL-6)。

BioVector? MMNK-1 is a highly differentiated, immortalized human cholangiocyte (bile duct epithelial) cell line.

• Origin: Human fetal liver.

• Cell Type: Immortalized normal cholangiocyte.

• Immortalization Method: Transduction with SV40 Large T antigen and human telomerase reverse transcriptase (hTERT).

• Morphology: Epithelial-like; exhibits highly differentiated characteristics of biliary cells.

• Markers: Positive for cholangiocyte-specific markers, including Cytokeratin-7 (CK-7) and Cytokeratin-19 (CK-19).

• Functional Traits:

  • Forms bile duct-like tubular structures in Matrigel assays.

  • Produces Interleukin-6 (IL-6) upon treatment with Lipopolysaccharide (LPS).

  • Forms bile duct-like structures when transplanted into immunodeficient mice.

  
  

• Reversibility: Contains LoxP sites flanking the SV40T gene, allowing for reversion to a non-immortal phenotype upon treatment with Cre recombinase.

• Medium: Typically cultured in specialized media such as modified MCDB-131 supplemented with EGM-2 SingleQuots (excluding ascorbic acid, which may be replaced by magnesium salt) and 2–10% FBS.

• Passaging: Recommend using Accutase or 0.25% Trypsin; use a 1:10 to 1:20 split ratio.

• Environment: 37°C in a 5% CO? atmosphere.

• Vessels: Collagen-coated dishes are generally not recommended by the original depositor as they may alter cell characteristics.
