BioVector? HIBEpiC (人肝內(nèi)膽管上皮細胞HIBECs) 是從健康人肝臟組織中分離的原代上皮細胞，主要襯于肝內(nèi)膽管樹表面。

• 全稱： 人肝內(nèi)膽管上皮細胞 (Human Intrahepatic Biliary Epithelial Cells)。

• 細胞類型： 原代上皮細胞。

• 組織來源： 人類正常肝臟組織。

• 細胞形態(tài)： 典型的鋪路石樣上皮細胞形態(tài)，貼壁生長。

• 質(zhì)量控制： 細胞通過 細胞角蛋白-18 (CK-18) 和 細胞角蛋白-19 (CK-19) 的免疫熒光鑒定。

• 病毒檢測： HIV-1、乙肝 (HBV)、丙肝 (HCV) 以及支原體、細菌、真菌檢測均為陰性。

• 推薦培養(yǎng)基： 上皮細胞專用培養(yǎng)基 (如 EpiCM)。也有研究使用 89% RPMI-1640 + 10% FBS + 1% 雙抗。

• 生長特性： 建議接種密度約為 5,000 cells/cm2。

• 擴增能力： 在官方推薦條件下，可保證進一步擴增約 10 次倍增 (Population Doublings)。

• 特殊要求： 培養(yǎng)瓶/皿通常需要預(yù)涂 聚左旋賴氨酸 (Poly-L-Lysine, PLL) 以促進細胞貼壁。

1. 潤洗： 吸除舊培養(yǎng)基，用 PBS 輕輕潤洗細胞。

2. 消化： 加入 0.25% Trypsin-EDTA 消化液。該細胞對消化較為敏感，嚴禁消化過度。

3. 終止： 當細胞間隙變大、細胞開始變圓時，立即加入含血清的完全培養(yǎng)基終止消化。

4. 離心： 1000 rpm (約 150g) 離心 3 分鐘，棄上清，用新鮮培養(yǎng)基重懸。

• 保存： 收到細胞后應(yīng)立即轉(zhuǎn)入 液氮 蒸氣層長期保存。嚴禁在 -80°C 長期存放，否則會導(dǎo)致細胞活力迅速下降。

• 運輸： 通常采用干冰運輸。

• 研究膽汁成分的調(diào)節(jié)與轉(zhuǎn)運。

• 研究膽汁性肝硬化、膽管癌及肝臟疾病的體外模型。

• 研究免疫反應(yīng)中的細胞因子和趨化因子分泌。

• Tissue Source: Human healthy liver.

• Morphology: Epithelial, typically growing as a tightly adherent monolayer.

• Characterization: Positive for markers cytokeratin-18 (CK-18) and cytokeratin-19 (CK-19).

• Viability: Generally guaranteed at ≥ 70% post-thaw; vials typically contain >500,000 viable cells.

• Growth Potential: Guaranteed for approximately 10–15 population doublings.

• Recommended Medium:Epithelial Cell Medium (EpiCM) or specialized Intra-Hepatic Biliary Epithelial Cell Growth Medium.

• Surface Coating: Culture vessels must be coated with Poly-L-Lysine (2 μg/cm2) to ensure proper attachment.

• Seeding Density: Recommended at 5,000 cells/cm2.

• Incubation Conditions: 37°C in a 5% CO? incubator.

• Subculturing: Perform when the culture reaches 90% confluency using 0.05% Trypsin/EDTA.

• Biosafety Level: BSL-1 or BSL-2 (depending on donor screening). Most suppliers test negative for HIV-1, HBV, HCV, and mycoplasma.

• Storage: Immediately transfer to liquid nitrogen vapor phase upon receipt. Do not store at -70°C as it will result in rapid loss of viability.

• Usage: For research use only; not for human or animal diagnostic or therapeutic procedures.
