Medium Supplement: Blasticidin S (at a final concentration of 4 μg/mL) must always be added to the culture medium to maintain the expression of the immortalization genes. Differentiation (Functional Assays): Temperature: To induce differentiation and form a functional barrier model, the culture temperature should be raised to 37°C. At this temperature, the tsSV40T antigen becomes unstable and loses its function, turning off immortalization signals.
