The GIST-48 cell line is an adherent human gastrointestinal stromal tumor (GIST) cell line established from a tumor that progressed during imatinib therapy. It is a key model for studying drug resistance due to its specific combination of KIT gene mutations. Key Characteristics Organism: Homo sapiens (Human) Disease: Gastrointestinal Stromal Tumor (GIST), specifically imatinib-resistant. Growth Properties: Adherent monolayer. Morphology: Spindle-shaped/epithelial-like cells. Key Mutations: The line is characterized by two KIT mutations: Primary Mutation: Homozygous KIT exon 11 V560D point mutation. Secondary (Resistance) Mutation: Heterozygous KIT exon 17 D820A missense mutation. Immunomarker: Expresses c-kit (CD117) and CD34. Derived Subline: The GIST48B subline was derived from GIST-48 after drug pressure, resulting in nearly undetectable KIT expression.
