培養(yǎng)基： RPMI 1640，補充 10% 熱滅活胎牛血清 (FBS) 和 2 mM L-谷氨酰胺。
孵育： 37°C，含 5% CO?。
傳代： 接種密度約為 0.5 x 10? 個細胞/毫升；每 2-3 天按 1:2 到 1:4 的比例傳代。最大密度約為 1.5 x 10? 個細胞/毫升。

關(guān)鍵標(biāo)志物和功能

陽性標(biāo)志物： HLA-DR、CD4、CD56、CD45RA、CD123 (IL-3Rα)。
陰性標(biāo)志物： CD3、CD11c（低水平）、CD14、CD16、CD19。
功能： 表達 TLR2、TLR4、TLR7 和 TLR9 的轉(zhuǎn)錄本，并在受到相應(yīng)配體（例如 CpG-ODNs、R848）刺激后產(chǎn)生 TNF-α、IFN-γ 和 IL-6 等細胞因子。

Culture Conditions

Medium: RPMI 1640 supplemented with 10% heat-inactivated Fetal Bovine Serum (FBS) and 2 mM L-glutamine.
Incubation: 37°C with 5% CO?.
Subculturing: Seed out at a density of approximately 0.5 x 10? cells/ml; split 1:2 to 1:4 every 2-3 days. The maximal density is around 1.5 x 10? cells/ml.
