The NTCC? cho-k1/aequorin/ga16 cell line is a genetically engineered Chinese Hamster Ovary (CHO-K1) cell line used as a biosensor to measure changes in intracellular calcium levels. It is used in research and drug discovery to study G-protein coupled receptors (GPCRs) and other signaling pathways.
Components and function
CHO-K1 host cells: A subclone of the original CHO cell line, CHO-K1 cells are a standard host for producing recombinant proteins and are easily transfected with foreign DNA. They are of Chinese hamster ovarian origin and require proline in their growth medium.
Aequorin: A photoprotein that emits light in the presence of calcium. In the cho-k1/aequorin/ga16 line, the aequorin gene is stably expressed and targeted to a specific cellular location, such as the mitochondria, to measure localized calcium changes.
Ga16: A chimeric G-protein alpha subunit that couples a wide range of receptors to the phospholipase C pathway, resulting in increased intracellular calcium. By co-expressing Ga16, the cell line can amplify the calcium response from various GPCRs.
GPCR screening: Because the Ga16 protein couples with many GPCRs, the cell line can be used to screen for compounds that activate these receptors, making it a versatile tool in early-stage drug discovery.
Calcium signaling studies: The aequorin reporter system allows for the detection of subtle or localized changes in intracellular calcium, which can provide insight into complex cellular signaling pathways.
Mitochondrial calcium dynamics: In versions where aequorin is targeted to the mitochondria, the cell line can be used to specifically investigate mitochondrial calcium influx and its role in cell death and metabolism.
