The temperature-sensitive replacement vector pBT2 contains the temperature-sensitive replicon of pE194 and the chloramphenicol resistance of pC194, both isolated from pTV1ts, the replicon and the β-lactamase gene of pBR322, and the multiple cloning site of pUC18. ， Thin lines respresent DNA derived from pBR322, thick lines from pTV1Ts. The location and orientation of resistance genes, origins of replication, and the T1 transcriptional terminator are marked by arrows.

Only relevant restriction sites are shown. In pBT1, the following restriction sites are available for cloning: EcoRI, HindIII, EcoRV, NheI, BamHI, PvuII. The multiple cloning site in pBT2 is derived from pUC18. Restriction sites indicated in the multiple cloning site are unique. In addition, EcoRV, NheI, and PvuII may be used for cloning in pBT2.
