established from the pleural effusion of an 80-year old patient with plasma cell leukemia in 1988. Cell line is IL-6 dependent and was described in the literature to carry an activating mutation of the N-RAS gene and del(1)(p22), del(7)(p13) and 14q+: t(11;14)(q13;q32) translocation

Morphology:
round cells of different sizes, growing singly or in small clumps in suspension;


Medium:
80% RPMI 1640 + 20% FBS + 10 ng/ml IL-6
Subculture:
seed out 0.5-1.0 x 106 cells/well in a 24-well plate or at 0.5 x 10^6 cells/ml in a small culture flask; maintain at 0.5-1.5 x 10^6 cells/ml, split 1:2 to 1:4 every 2-3 days. After thawing growth may be delayed

at 37 °C with 5% CO2
Doubling time:
about 30-50 hours
Harvest:
maximal density of ca. 2 x 10^6 cells/ml
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
