The library vector, pLN-CD90-CMV-gp130-ccdB, was constructed from pLNCX2 (Clontech) by first destroying the EcoRI site at 5991 and then cloning the CD90 gene into the EcoRI (1471) and NaeI (2143) sites. MfeI (compatible with EcoRI, both sites destroyed after ligation) and NaeI sites were created by PCR with primers CD90-MfeI and CD90-NaeI. A gp130-ccdB fragment was cloned into the XhoI and ClaI sites by PCR amplification from pBG1 [20] with primers gp130-XhoI and ccdB-ClaI. This fragment carries a unique EcoRI site at position 3284 that is used for cDNA cloning.
